چکیده

An intracellular transport of proline and glycine was examined by using Ehrlich ascites carcinoma cells. The pH optima and the effect of heat inactivation on the intracellular uptake were found to be completely the same for both proline and glycine. The Michaelis type constant of glycine transport was the same as its inhibition constant to the proline transport. Moreover the Michaelis type constant of proline transport was almost the same as its inhibition constant to the glycine transport. The inhibition constants of hydroxyproline, and also of eveloleucine, were proved to be the same as those of both proline and glycine transport. From these results the existence of a transport system common to both imino acids and glycine was emphasized.

Transport of amino acids in renal brush border membrane vesicles. Uptake of L-proline.

The findings (a) stereospecific counter transport; (b) equilibrium uptake of L-proline decreased with increasing medium osmolarity; and (c) L-proline and Na+ were taken up into identical intravesicular spaces, indicate that uptake of L-proline by rabbit renal brush border membranes represents transport into membrane vesicles, not surface binding to the membrane. An Na+ gradient between extraves...

Renal imino acid and glycine transport system ontogeny and involvement in developmental iminoglycinuria.

Renal maturation occurs post-natally in many species and reabsorption capacity at birth can vary substantially from the mature kidney. However, little is known regarding the maturation of amino acid transport mechanisms, despite the well-known physiological state of developmental iminoglycinuria. Commonly seen during early infancy, developmental iminoglycinuria is a transient version of the per...

Iminoglycinuria and hyperglycinuria are discrete human phenotypes resulting from complex mutations in proline and glycine transporters.

Iminoglycinuria (IG) is an autosomal recessive abnormality of renal transport of glycine and the imino acids proline and hydroxyproline, but the specific genetic defect(s) have not been determined. Similarly, although the related disorder hyperglycinuria (HG) without iminoaciduria has been attributed to heterozygosity of a putative defective glycine, proline, and hydroxyproline transporter, con...

Chloride dependent amino acid transport in the human small intestine.

Carriers of beta amino acids and imino acids in the small intestine of rabbits and guinea pigs are chloride dependent, and a cotransport of chloride, sodium, and 2-methyl-aminoisobutyric acid has been shown. This study examines the chloride dependence of amino acid transport in the human small intestine. The steady state tissue uptake of amino acids, given as the ratio between substrate concent...

Renal tubular transport of proline, hydroxyproline, and glycine. II. Hydroxy-l-proline as substrate and as inhibitor in vivo.

Renal tubular absorption of the free amino acids L-proline, hydroxy-L-proline, and glycine in man is accomplished by a transport system exhibiting preference for this group of amino acids (1, 2). The characteristics of saturability (for L-proline transport) and selective inhibition between the constituent substrates have been demonstrated for this system in man (1). Studies in vivo in the rat (...
